Bacteria are very small and cannot seen with the naked eye. To see them we need a microscope that magnifies about 500 times to observe them.
If a bacteria cell or a mold spore is allowed to grow on suitable food or solid media, it will reproduce
rapidly into a colony consisting of millions of cells. The colony
is visible to the naked eye. The media is supplying the bacteria with food. If the food is optimal these bacteria will grow fast.
That is why we call this trademark "Bactophyl" meaning "We love bacteria".
Bacteria and Fungi come under different categories; the former one is the prokaryotic cell while the latter one is Eukaryotic cells.
Apart from this, there are many differences between them like bacteria need a host to live, and they can be autotrophs as well as heterotrophs,
whereas fungi grow their own and are heterotrophs which depend on others for their food.
Bacteria do not have nuclear membrane enclosing nucleus, while Fungi (eukaryotes) have well- defined nucleus surrounded by the nuclear membrane.

Agar-Agar

Agar, or agar-agar, is a jelly-like substance, obtained from red algae. Agar is a mixture of two components: the linear polysaccharide agarose, and a heterogeneous mixture of smaller molecules called agaropectin.
Agar in microbiology is used to give the liquid components solidity and to let the micro-organisms grow on the surface.
Nutrient Agar is a general purpose, nutrient medium used for the cultivation of microbes supporting growth of a wide range of non-fastidious organisms.
Nutrient agar is popular because it can grow a variety of types of bacteria and fungi, and contains many nutrients needed for the bacterial growth.

0.5% Peptone - this provides organic nitrogen
0.3% beef extract/yeast extract - the water-soluble content of these contribute vitamins, carbohydrates, nitrogen, and salts
1.5% agar - this gives the mixture solidity
0.5% Sodium Chloride - this gives the mixture proportions similar to those found in the cytoplasm of most organisms
distilled water - water serves as a transport medium for the agar's various substances
pH adjusted to neutral (6.8) at 25 °C (77 °F).
Yeast extract is produced commercially by heating a suspension of yeast; the enzymes in the yeast cell then degrade the cell wall.
The result has more concentrated flavor and a different texture. Yeast extract has a protein content of about 50%, of which about 20% is glutathione,
6% is nucleic acid. It is rich in 18 kinds of amino acids, functional peptides glutathione, dextran, mannan, trehalose,
flavoring nucleotide, B vitamins, biotin, trace elements and volatile aromatic compounds and other components.
Yeast Mold Agar is a medium used for the isolation and cultivation of yeast and molds.
Mold is a type of fungus that grows in multicellular filaments called hyphae.
These tubular branches have multiple, genetically identical nuclei, yet form a single organism, known as a colony.
In contrast, yeast is a type of fungus that grows as a single cell.
Peptone and Malt extract provides the carbon, protein and nutrient sources required for the growth of microorganisms.

Potato dextrose agar

Potato Dextrose Agar (PDA) is used for the cultivation of fungi.
Potato Dextrose Agar (PDA) is a general purpose medium for yeasts and molds that can be supplemented with acid or antibiotics to inhibit bacterial growth.
It is recommended for plate count methods for foods, dairy products and testing cosmetics.
PDA can be used for growing clinically significant yeast and molds. The nutritionally rich base (potato infusion) encourages mold sporulation and pigment production in some dermatophytes.
Principle of Potato Dextrose Agar (PDA)
Potato Dextrose Agar is composed of dehydrated Potato Infusion and Dextrose that encourage luxuriant fungal growth.
Agar is added as the solidifying agent. Many standard procedures use a specified amount of sterile tartaric acid (10%) to lower the pH of this medium to 3.5 +/- 0.1, inhibiting bacterial growth.
Chloramphenicol acts as a selective agent to inhibit bacterial overgrowth of competing microorganisms from mixed specimens, while permitting the selective isolation of fungi.

Czapek Dox agar

Czapek medium, also called Czapek's agar (CZA)or Czapek-Dox medium, is a growth medium for propagating fungi and other organisms in a laboratory. It was named after its inventors, Czech botanist Friedrich Johann Franz Czapek (1868 - 1921) and American chemist Arthur Wayland Dox (1882 - 1954). The medium contains sucrose as the sole source of carbon and nitrate as the only inorganic source of nitrogen. The medium is useful in a variety of microbiological procedures, including fungi and mildew resistance tests and soil microbiology testing. Czapek-Dox Agar produces luxuriant growth of most saprophytic Aspergilli causing the organisms to produce characteristic mycelia and conidia. Czapek-Dox Agar is recommended in Standard Methods for the Examination of Water and Wastewater for the isolation of Aspergillus, Penicillium, Paecilomyces, and other types of fungi with similar physiological requirements. Simply said it works well for many saprophytic fungi and soil bacteria.
